Functionally, GLRX3 acts together with BOLA2 as a cytosolic iron-sulfur (Fe-S) cluster assembly factor that facilitates [2Fe-2S] cluster insertion into a subset of cytosolic proteins, and acts as a critical negative regulator of cardiac hypertrophy and a positive inotropic regulator. It is required for hemoglobin maturation and lacks thioredoxin catalytic activity. The protein forms a homodimer independent of 2Fe-2S clusters and a heterotrimeric complex composed of two BOLA2 molecules and one GLRX3, linked by [2Fe-2S] clusters.
