Glutamine synthetase (GS) is the ATP-dependent enzyme that condenses glutamate and free ammonia into glutamine, supporting nitrogen balance, hepatic ammonia detoxification, astrocytic glutamate recycling in the brain, and the glutamine-addiction phenotype observed in many cancer cell lines. This microplate immunoassay from Assay Genie provides quantitative measurement of endogenous mouse GS protein across tissue homogenates, cell lysates, and cell culture supernatants from murine experimental systems.

The kit ships with pre-coated plate wells, calibrated recombinant mouse GS standards, detection antibodies, and substrate buffers sufficient for 96 individual reactions. An absorbance-based readout generates a standard curve that interpolates unknown sample concentrations and is compatible with standard laboratory microplate readers. Each component has been validated on mouse-derived matrices to maintain quantitative recovery, and the workflow supports both endpoint and serial sampling study designs.

Reagents should be stored refrigerated upon receipt and protected from light; aliquot the standard curve protein if partial plates will be run across multiple sessions to avoid freeze-thaw cycling of the calibrator. The kit is configured for in vitro research use only and is commonly deployed alongside western blot validation when assessing GS expression in engineered mouse strains, hepatic injury models, and glioblastoma or hepatocellular carcinoma xenograft studies. Investigators investigating glutamate-glutamine cycling in neural tissue or c-Myc-driven metabolic rewiring in tumor models routinely incorporate this reagent as part of their workflow.
