Description

From Assay Genie, this Mouse GIP (Gastric Inhibitory Polypeptide) ELISA Kit is a sandwich ELISA built on a double-antibody configuration for the quantitative determination of GIP in mouse samples. Colorimetric detection is achieved with TMB substrate. Gastric inhibitory polypeptide is described as a key hormone involved in the regulation of glucose metabolism and insulin secretion, with a role in the gut-brain axis influencing food intake and energy balance. Dysregulation of GIP has been linked to obesity, diabetes, and metabolic disorders, making it a biomarker of interest for metabolic disease research.

The kit reports a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. Compatible sample matrices include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity for serum (n=5) at 1:2, 1:4, and 1:8 dilutions is 88–98%, 90–100%, and 84–93%; EDTA plasma (n=5) shows 86–99%, 86–98%, and 80–91%; heparin plasma (n=5) shows 87–105%, 89–101%, and 83–100%. Recovery is 85–103% (average 95%) in serum (n=5), 92–105% (average 97%) in EDTA plasma, and 87–105% (average 93%) in heparin plasma. Storage is 2–8°C for 12 months.

The workflow uses a biotin-labeled detection antibody incubated at 37°C for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, TMB substrate for color development, and absorbance measurement at 450 nm. Sample preparation guidance includes clotting and centrifugation of serum at 1000 × g for 20 minutes, collection of plasma with anticoagulant and centrifugation at 1000 × g for 15 minutes at 2–8°C, and centrifugation of cell culture supernatant at 2500 rpm for 5 minutes. Provided components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual.
