Description

Designed by Assay Genie for the precise measurement of gastrin levels in mouse serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, this competitive ELISA is coated with antigen and read colorimetrically at 450 nm. The assay has a sensitivity of 0.188 pg/mL and a working range of 0.313–20 pg/mL. Gastrin is a pivotal hormone involved in regulating gastric acid secretion and promoting the growth of gastric mucosa, and its dysregulation has been linked to gastrointestinal disorders such as gastritis, peptic ulcers, and gastric cancer, underscoring its value as a biomarker for studying these conditions.

The protocol begins with equilibrating TMB substrate for 30 minutes at room temperature, preparing standards and samples at a minimum 1:2 dilution, and pre-washing the plate twice. Each 50 µL of standard or sample is combined with 50 µL of biotin-labeled antibody, mixed gently, and incubated at 37°C for 45 minutes. The plate is washed five times with wash buffer, TMB substrate is added, and colour is developed in the dark at 37°C for 10–20 minutes. After adding stop solution (colour changes from blue to yellow), absorbance is read at 450 nm using a microplate reader. The kit is stored sealed at 2–8°C and should not be cryopreserved.
