Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative detection of mouse Gamma-secretase subunit APH-1A (Aph1a), also known by the synonyms Aph-1alpha and APH-1a. The assay reacts with mouse samples and is validated for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with specificity for both natural and recombinant mouse Gamma-secretase subunit APH-1A.

Aph1a is a non-catalytic subunit of the gamma-secretase complex, an endoprotease that catalyses the intramembrane cleavage of integral membrane proteins such as Notch receptors and the beta-amyloid precursor protein (APP). It is required for normal gamma-secretase assembly and participates in Notch and Wnt signalling cascades, and dysregulation of gamma-secretase activity has been linked to Alzheimer's disease and cancer. The protein localises predominantly to the endoplasmic reticulum membrane and Golgi stack membrane, and the kit is grouped under the Neurosciences research area. Performance specifications include a detection range of 1.56-100ng/mL, a sensitivity of 0.81ng/mL, an intra-assay CV of 6.2% and an inter-assay CV of 7.9%. Linearity has been demonstrated in serum (109-118%, 111-120%, 91-100%, 105-116% across dilutions 1:2 to 1:16), EDTA plasma (89-99%, 96-106%, 86-96%, 89-89%) and heparin plasma (111-120%, 87-99%, 86-99%, 92-104%), with average recovery of 101% in serum (range 95-107%) and 103% in plasma (range 97-109%). The kit is for research use only.
