Description

This sandwich ELISA kit from Assay Genie is designed to measure mouse Gamma-glutamyltranspeptidase 1 (Ggt1) in serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids. The assay uses a colorimetric detection method and is specific for natural and recombinant mouse glutathione hydrolase 1 proenzyme.

The kit has a detection range of 0.156-10 ng/mL with a sensitivity of 0.08 ng/mL. Reported intra-assay CV is 5.1% and inter-assay CV is 7.6%. Recovery in serum averages 97% (range 91-103%) and in plasma averages 99% (range 93-105%). Linearity values for serum at dilutions of 1:2, 1:4, 1:8, and 1:16 were 99-109%, 102-111%, 94-104%, and 115-125%, with comparable ranges reported for EDTA and heparin plasma matrices.

Ggt1 is a plasma membrane-associated enzyme that cleaves the gamma-glutamyl bond of extracellular glutathione and related conjugates, releasing free glutamate and the dipeptide cysteinyl-glycine. The protein is part of the cellular antioxidant defense mechanism, supplies a local cysteine source, contributes to maintenance of intracellular GSH, and indirectly regulates multiple aspects of skeletal biology. The functional enzyme is a heterodimer of heavy and light chains, with the active site in the light chain, and is anchored as a single-pass type II membrane protein (UniProt Q60928, NCBI Gene ID 14598). Synonyms include gamma-glutamyltransferase 1, leukotriene-C4 hydrolase, GGT 1, CD224, Ggt, and Ggtp. The kit is for research use only.
