The Mouse Gamma-Endorphin (gEP) ELISA Kit from Assay Genie uses a competitive immunoassay format in which gamma-endorphin present in the test sample competes with a reference conjugate for binding to a specific antibody immobilized on the microplate. Gamma-endorphin (β-endorphin-(1-17), γ-EP) is a 17-residue endogenous opioid neuropeptide generated by prohormone convertase cleavage of proopiomelanocortin (POMC). The peptide shares its N-terminal Tyr-Gly-Gly-Phe-Met sequence with the larger β-endorphin-(1-31) but lacks the C-terminal extension, making it a useful marker of post-translational POMC processing in murine neuroendocrine tissues.

Researchers employ this in vitro assay in neuroscience and endocrinology studies of opioid peptide biology, hypothalamic-pituitary stress-axis reactivity, nociceptive modulation, and behavioral correlates of neuropeptide release in transgenic or pharmacologically treated mice. Validated sample types include mouse serum, lithium-heparin-treated plasma, and freshly prepared brain or pituitary tissue homogenates. Reported performance characteristics include a standard detection range of approximately 25 to 1000 pg/mL, sensitivity near 5 pg/mL, intra-assay precision below 10% CV, and inter-assay precision below 12% CV. The recommended sample volume is 50 µL per well. Unopened kit components are kept refrigerated; reconstituted standards and working aliquots are best stored frozen and protected from repeated freeze-thaw cycles to preserve peptide integrity.
