Verified performance characteristics for this Assay Genie sandwich ELISA format include a stated shelf life of 12 months from manufacture when the sealed kit is held at 2 to 8 degrees C, with a measured loss of activity below 5% across that period under appropriate storage. Spike-and-recovery studies on the calibration range give typical recoveries of 80 to 102% in murine serum (average 91%), 81 to 100% in EDTA plasma (average 90%) and 80 to 89% in heparin plasma (average 84%), and linearity is maintained across 1:2 to 1:16 dilutions of spiked serum, EDTA plasma and heparin plasma, with calculated concentrations recovering between roughly 80 and 104% of expected values.

Precision data supplied with the kit document intra-assay coefficients of variation below 10% and inter-assay CVs below 12% when three serum pools spanning the calibration curve are tested in twenty replicates on a single plate or across three plates, respectively. Specificity testing indicates that the antibody pair recognizes GAL7 with no significant cross-reactivity with homologous galectin family members, so signals can be attributed to the LGALS7 gene product rather than to galectin-1, galectin-3 or related beta-galactoside-binding lectins present in the same specimen.

The full detection protocol requires approximately 4 hours of hands-on and incubation time once reagents have equilibrated to room temperature, excluding sample preparation. Recommended sample volumes differ by matrix: 50 uL for serum, plasma and other liquid samples, and 100 uL for cell culture supernatant or clarified tissue lysate. Tissue samples are typically homogenized in cold lysis buffer supplemented with protease inhibitors and clarified by centrifugation at 10,000 x g for 10 minutes at 4 degrees C before loading. The kit is intended for research use only.
