Description

This Sandwich ELISA (Double Antigen) from Assay Genie measures mouse Glutamate Decarboxylase Autoantibody (GADAb) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Colorimetric readout is taken at 450 nm after TMB substrate development. Reported sensitivity is 0.938 ng/mL and the standard range is 1.563–100 ng/mL. The kit is stored sealed at 2–8°C and should not be cryopreserved.

Linearity (n=5) at 1:2, 1:4 and 1:8 dilutions: serum 81–96%, 86–97%, 80–100%; EDTA plasma 89–105%, 86–98%, 83–96%; heparin plasma 86–103%, 84–100%, 85–98%. Spike recovery: serum 86–100% (average 91%), EDTA plasma 87–102% (average 98%), heparin plasma 86–105% (average 93%). The protocol runs primary incubation at 37°C for 90 minutes, biotin-labelled antigen for 60 minutes, HRP-streptavidin for 30 minutes, then TMB for 10–20 minutes in the dark; stop solution is added and absorbance is read at 450 nm. Serum samples are clotted and centrifuged at 1000 × g for 20 minutes; plasma in anticoagulant tubes is centrifuged at 1000 × g for 15 minutes at 2–8°C.

GADAb targets the enzyme glutamate decarboxylase, which functions in inhibitory neurotransmission. Abnormal GADAb levels have been linked to autoimmune diseases such as type 1 diabetes and to neurological disorders including epilepsy and stiff person syndrome.
