The Assay Genie Mouse G6PC immunoassay targets the endoplasmic reticulum-embedded catalytic subunit encoded by the G6pc1 gene, also annotated as G6PT, GSD1a, and historically linked to von Gierke glycogen storage disease type I. Because this 40 kDa transmembrane protein is the terminal enzyme in both gluconeogenesis and glycogenolysis, its expression serves as a quantitative readout of hepatic glucose output capacity. Studies employing this kit commonly pair the readout with fasting insulin, pyruvate tolerance, and PEPCK expression data to characterize the gluconeogenic axis in diet-induced obese, streptozotocin-treated, and liver-specific knockout mouse models.
Each kit is supplied with a pre-coated antibody plate, biotinylated detection reagent, streptavidin-HRP conjugate, lyophilized calibrator stock, wash concentrate, substrate solution, and stop reagent in individually sealed vials. Reconstituted standards and diluted samples should be kept frozen and protected from repeated freeze-thaw cycles to preserve epitope integrity. Working dilutions of the biotinylated antibody and streptavidin-HRP conjugate are prepared fresh before each run and discarded afterward. The complete incubation sequence requires approximately four hours at room temperature, with 37 degrees C steps during sample binding and detection incubations.
Recovery in fortified mouse serum has been demonstrated between 85 and 105 percent, with parallelism preserved at dilutions from 1:2 through 1:16. The assay is calibrated against recombinant catalytic subunit standard and exhibits no appreciable cross-reactivity with paralogous glucose-6-phosphatase-related proteins. Researchers apply it to liver, kidney, and small intestine homogenates, as well as to cultured hepatocyte supernatant fractions, to monitor catalytic subunit abundance across genetic, dietary, and pharmacological interventions relevant to type 2 diabetes and non-alcoholic fatty liver disease research.
