Beyond the core specifications already described, the Assay Genie Mouse FLT1/VEGFR1 ELISA Kit is built on a double-antibody sandwich format in which a pre-coated anti-VEGFR1 capture antibody pairs with a biotinylated detection antibody and an HRP-Streptavidin conjugate, with TMB as the chromogenic substrate. The target is recognized at the mouse Flt1 locus (UniProt P35969), and the kit's aliases include Fms-Related Tyrosine Kinase 1, Vascular Permeability Factor Receptor, Fms-like tyrosine kinase 1, and Tyrosine-protein kinase receptor FLT. Performance data show no significant cross-reactivity with analogous receptors, so readouts reflect VEGFR1 specifically rather than related VEGF-binding proteins.

Serum samples are typically clotted and centrifuged at 1000 x g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 x g for 15 minutes; cell culture supernatant is clarified at 2500 rpm for 5 minutes before loading onto the pre-coated microwell strips. The full assay run, excluding equilibration and sample prep, takes roughly 3.5 to 4 hours, with sequential incubations and wash cycles executed on a standard microplate reader set to 450 nm. Because VEGFR1 binds VEGF-A, VEGF-B, and placental growth factor, this ELISA is frequently paired with VEGF ligand measurements in studies of tumor angiogenesis, retinal neovascularization, pre-eclampsia modeling, and post-ischemic vascular remodeling in murine systems. Reagents that remain after first use should be returned to refrigerated storage at 4 degrees C for short-term use, or aliquoted and frozen at -20 degrees C for longer-term stability to limit freeze-thaw loss of antibody activity.
