Mouse FGF21 ELISA Kit

From Assay Genie, this colorimetric Sandwich ELISA Kit is designed for the precise measurement of FGF21 — a member of the fibroblast growth factor family involved in regulating glucose and lipid metabolism, as well as energy homeostasis — in mouse samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It uses a double-antibody Sandwich ELISA format with a sensitivity of 18.75 pg/mL and a measurable range of 31.2–2000 pg/mL.

Validated linearity values for serum (n=5) range from 88–102% at 1:2, 88–101% at 1:4, and 94–98% at 1:8. EDTA plasma (n=5) shows 82–97%, 83–101%, and 84–99% at the same dilutions, while heparin plasma (n=5) shows 82–99%, 92–97%, and 80–98%. Recovery averages are 92% for serum (range 85–103%), 97% for EDTA plasma (range 90–103%), and 95% for heparin plasma (range 86–105%).

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution. The kit should be stored at 2–8°C for 12 months.
