Mouse Fgf15/Fibroblast growth factor 15 ELISA Kit

This sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of mouse Fibroblast Growth Factor 15 (FGF15), a regulator of bile acid synthesis and metabolism that has been implicated in non-alcoholic fatty liver disease (NAFLD) and metabolic syndrome. The assay uses a double-antibody sandwich format with colorimetric detection and is intended for use with mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported sensitivity is 2.813 pg/mL with an assay range of 4.688–300 pg/mL. The kit is stored at 2–8°C for 12 months. Spiked recovery in mouse samples is reported as 85–99% (average 94%) for serum, 92–103% (average 99%) for EDTA plasma, and 88–99% (average 94%) for heparin plasma. Linearity across 1:2, 1:4 and 1:8 dilutions falls within 80–103% depending on the matrix.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition. Recommended sample preparation includes clotting and centrifugation of serum at 1000 × g for 20 minutes, collection of plasma in anticoagulant tubes followed by centrifugation at 1000 × g for 15 minutes at 2–8°C, and homogenisation of tissue in PBS with protease inhibitors.
