Mouse Fatty Acid Synthase/FASN ELISA Kit

A sandwich ELISA, double-antibody kit from Assay Genie enables quantitative measurement of mouse Fatty Acid Synthase (FASN; synonyms FAS, OA-519, SDR27X1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a biotinylated detection antibody with HRP-Streptavidin and TMB substrate, with absorbance read at 450 nm after a stop reaction.

Recovery values were 92–105% (average 99%) for serum, 89–98% (average 94%) for EDTA plasma and 94–102% (average 97%) for heparin plasma. Linearity across 1:2, 1:4 and 1:8 dilutions was reported as 86–104% / 89–100% / 83–100% for serum, 87–96% / 86–97% / 81–97% for EDTA plasma and 87–105% / 83–96% / 83–99% for heparin plasma. The kit is intended for storage at 2–8°C for 12 months and falls within the Metabolism research area.

The procedure comprises six steps: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding at 37°C for 60 minutes; HRP-Streptavidin binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and stop reaction followed by absorbance measurement at 450 nm.
