Description

Designed for the accurate quantification of Fatty Acid Binding Protein 6 (FABP6) in mouse samples, this colorimetric Sandwich ELISA kit from Assay Genie offers high sensitivity and specificity for research applications spanning lipid metabolism and bile acid regulation. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a working sensitivity of 46.875 pg/mL and a detection range of 78.125–5000 pg/mL.

FABP6, also known by synonyms including gastrotropin, I-BABP, ILBP, and ileal bile acid-binding protein, is a key regulator of enterohepatic bile acid metabolism whose dysregulation has been linked to liver disease, inflammatory bowel disease, and metabolic disorders. The double-antibody sandwich assay is supplied with a pre-coated dismountable microplate, lyophilized standard, biotin-labeled detection antibody, HRP-Streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, wash buffer, stop solution, plate sealer, and technical manual. All components should be stored at 2–8°C and remain stable for 12 months under these conditions.

The protocol proceeds as follows: equilibrate reagents and TMB substrate to room temperature, then incubate standards and samples on the pre-coated plate at 37°C for 90 minutes. Add the biotin-labeled detection antibody and incubate for 60 minutes at 37°C, followed by HRP-Streptavidin for 30 minutes. Develop colour with TMB substrate in the dark for 10–20 minutes, stop the reaction, and measure absorbance at 450 nm. Reported linearity covers serum (88–102% at 1:2, 85–104% at 1:4, 86–102% at 1:8), EDTA plasma (86–98%, 85–101%, 82–96%), and heparin plasma (83–97%, 85–100%, 80–95%). Mean recovery is 92% for serum, 95% for EDTA plasma, and 96% for heparin plasma.
