Description

A sandwich-format ELISA from Assay Genie for the quantitative detection of mouse Eukaryotic translation initiation factor 2-alpha kinase 3 (Eif2ak3), also referred to as PRKR-like endoplasmic reticulum kinase, Pancreatic eIF2-alpha kinase, Pek, or Perk. The kit reacts with mouse samples and is suitable for serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids. Reported specificity covers natural and recombinant mouse EIF2AK3.

Reported performance characteristics include a detection range of 0.312-20ng/mL, a sensitivity of 0.161ng/mL, an intra-assay CV of 6.9%, and an inter-assay CV of 9.0%. Recovery is reported as 81% (range 80-87%) for serum and 83% (range 80-89%) for plasma. Linearity has been assessed across serial dilutions of 1:2, 1:4, 1:8, and 1:16 in serum, EDTA plasma, and heparin plasma, with percentage ranges reported for each matrix.

EIF2AK3 is a metabolic-stress sensing protein kinase that phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (eIF-2-alpha/EIF2S1) on 'Ser-52' during the unfolded protein response and in response to low amino acid availability. It serves as a critical effector of unfolded protein response-induced G1 growth arrest through loss of cyclin-D1 and contributes to control of mitochondrial morphology and function. Dysfunction of EIF2AK3 has been linked to diabetes, neurodegenerative disorders, and certain cancers. The kit is listed for research use only, and the associated research area is Neurosciences.
