Mouse Endothelin-2/ET-2 ELISA Kit

A sandwich ELISA using a double-antibody format from Assay Genie enables quantitative measurement of Mouse Endothelin-2/ET-2 in mouse-derived samples. The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with reagents stored at 2-8°C for 12 months. Reported performance includes a sensitivity of 0.469 pg/mL and a standard range of 0.781-50 pg/mL. The assay targets the Endothelin-2 peptide, listed under the synonyms Edn2, ET-2, Preproendothelin-2 and PPET2 (UniProt P22389), and uses a colorimetric detection method.

The procedure begins with equilibration of reagents and TMB substrate to room temperature, followed by loading of standards, samples and blanks onto the pre-coated plate and a primary incubation at 37°C for 90 minutes. A biotin-labelled detection antibody is added and incubated at 37°C for 60 minutes, then HRP-Streptavidin (SABC) is applied for 30 minutes at 37°C. Colour is developed with TMB substrate in the dark for 10–20 minutes, the reaction is stopped, and absorbance is measured at 450 nm.

Linearity data are reported for serum (n=5) at 89-99% (1:2), 82-100% (1:4) and 84-100% (1:8); EDTA plasma (n=5) at 89-99% (1:2), 84-100% (1:4) and 81-97% (1:8); and heparin plasma (n=5) at 86-100% (1:2), 83-94% (1:4) and 87-96% (1:8). Recovery ranges and averages are: serum (n=5) 92-100%, average 94%; EDTA plasma (n=5) 92-101%, average 97%; heparin plasma (n=5) 88-105%, average 98%.
