Mouse Endothelin-1 ELISA Kit

Designed by Assay Genie for research applications, this sandwich-format ELISA kit enables quantitative measurement of endothelin-1 in mouse serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody assay uses a colourimetric TMB substrate read at 450 nm. Endothelin-1 is implicated in vasoconstriction and cell growth, with relevance to cardiovascular disease, hypertension and kidney disorders.

Sensitivity is reported at 0.625 pg/mL with a working range of 1.25–80 pg/mL. Linearity was evaluated across 1:2, 1:4 and 1:8 dilutions: serum (n=5) returned 85–103%, 90–103% and 91–102%; EDTA plasma (n=5) returned 85–100%, 83–98% and 84–96%; heparin plasma (n=5) returned 82–94%, 89–99% and 83–100%. Spike-and-recovery averages were 95% for serum, 95% for EDTA plasma and 98% for heparin plasma, with corresponding recovery ranges of 87–102%, 86–101% and 93–103%. Unopened kit components are stored at 2–8°C for 12 months.

The procedure covers reagent and plate preparation, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes at 37°C, HRP-streptavidin (SABC) binding for 30 minutes at 37°C, TMB colour development in the dark for 10–20 minutes, and a stop reaction with immediate absorbance measurement at 450 nm.
