Linearity in serum (N=5) ranges from 82-94% at 1:2 to 80-90% at 1:16, in EDTA plasma from 84-97% to 92-100%, and in heparin plasma from 90-100% to 96-106%. Recovery averages 86% in serum (range 80-92%) and 88% in plasma (range 82-94%). The assay specifically recognises natural and recombinant mouse E3 SUMO-protein ligase PIAS3 (UniProt O54714), a monomeric protein localised to the cytoplasm, nucleus, and nuclear speckles.

PIAS3 belongs to the protein inhibitor of activated STAT (PIAS) family and functions as an E3-type SUMO ligase and transcriptional coregulator, modulating cellular processes including the STAT pathway and steroid hormone signalling. It acts as a repressor of STAT3 DNA-binding and suppresses cell growth, with dysregulation linked to cancer, inflammation, and immune disorders, supporting its relevance as a research target for disease mechanisms and therapeutic investigation.
