Description

Designed for the precise measurement of MSH2 levels in mouse serum, plasma, and cell culture supernatants, this Sandwich ELISA from Assay Genie offers high sensitivity and specificity for a range of research applications. MSH2 is a critical protein involved in DNA mismatch repair, ensuring the proper functioning of genetic material and maintaining genomic stability. Defects in MSH2 have been linked to hereditary nonpolyposis colorectal cancer (HNPCC) and other cancers, highlighting its importance as a biomarker for cancer research.

The assay delivers a detection range of 78-5000pg/mL with a sensitivity of 39pg/mL. Reproducibility is supported by an intra-assay CV of 4.4% and an inter-assay CV of 6.9%. Recovery averages 88% (range 82-94%) for serum and 90% (range 84-96%) for plasma. Specificity is confirmed for natural and recombinant mouse DNA mismatch repair protein Msh2 (UniProt P43247).

Functionally, MSH2 forms two heterodimers, MutS alpha (MSH2-MSH6) and MutS beta (MSH2-MSH3), which bind to DNA mismatches and initiate repair. MutS alpha recognises single base mismatches and dinucleotide insertion-deletion loops, while MutS beta recognises larger insertion-deletion loops. The protein localises to the nucleus and is a component of the BRCA1-associated genome surveillance complex (BASC). The kit is intended for research use only, supporting studies of DNA repair mechanisms, disease pathways, and the design of targeted treatments in mouse models.
