Description

From Assay Genie, this sandwich ELISA kit uses a double-antibody format with colorimetric detection to quantify Dihydrolipoamide Acetyltransferase (DLAT), also known as the Pyruvate Dehydrogenase Complex (PDC) E2 subunit, in mouse samples. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples.

Reported linearity across 1:2 to 1:8 dilutions is 86–104%, 87–105%, and 87–104% for serum (n=5); 82–99%, 85–101%, and 83–92% for EDTA plasma (n=5); and 85–96%, 82–92%, and 85–100% for heparin plasma (n=5). Spike-and-recovery averages were 97% for serum (range 90–105%), 98% for EDTA plasma (range 87–103%), and 98% for heparin plasma (range 93–104%). Unopened kits are stored at 2–8°C and remain stable for 12 months.

DLAT is a key enzyme of the pyruvate dehydrogenase complex, essential for the conversion of pyruvate to acetyl-CoA in the citric acid cycle, and its dysregulation has been implicated in metabolic disorders such as diabetes and metabolic syndrome, making this kit a relevant tool for metabolism research. The protocol uses a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-Streptavidin binding step, 10–20 minutes of TMB colour development in the dark, and immediate absorbance reading at 450 nm after stop solution is added.
