Description

Designed for the accurate detection of mouse diamine oxidase (DAO) levels, this Sandwich ELISA from Assay Genie uses a double-antibody format with a colorimetric readout measured at 450 nm. The assay is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, supporting research into histamine metabolism, allergic responses, gastrointestinal disorders, and histamine intolerance in mouse models.

Sensitivity is reported at 0.938 ng/mL with a working range of 1.563–100 ng/mL, and kits are stored at 2–8°C for up to 12 months. Validation data include linearity across serial dilutions — Serum: 85–103% (1:2), 85–103% (1:4), 94–99% (1:8); EDTA Plasma: 82–101% / 84–98% / 87–98%; Heparin Plasma: 86–100% / 81–99% / 82–96% — and recovery values of Serum 89–105% (average 98%), EDTA Plasma 86–100% (average 94%), and Heparin Plasma 91–101% (average 96%).

The workflow follows a standard sandwich protocol: equilibrate reagents and TMB substrate, incubate standards and samples on the pre-coated plate at 37°C for 90 minutes, add biotin-labelled detection antibody for 60 minutes, then HRP-Streptavidin (SABC) for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, stop the reaction, and read absorbance at 450 nm. Serum samples are allowed to clot and centrifuged at 1000 × g for 20 minutes, while plasma is collected using anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C.
