Description

A colorimetric sandwich ELISA kit from Assay Genie designed for the quantitative measurement of mouse delta-1-pyrroline-5-carboxylate dehydrogenase, mitochondrial (ALDH4A1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay is reactive with mouse samples and detects natural and recombinant mouse ALDH4A1. Storage: -20°C; shipping at +4°C.

ALDH4A1 is a mitochondrial matrix enzyme that catalyses the irreversible conversion of delta-1-pyrroline-5-carboxylate (P5C), derived from proline or ornithine, to glutamate, a necessary step in the pathway interconnecting the urea and tricarboxylic acid cycles. The preferred substrate is glutamic gamma-semialdehyde; other substrates include succinic, glutaric and adipic semialdehydes. The protein forms a homodimer and is involved in proline catabolism, amino acid metabolism, energy production and the oxidative stress response. Dysregulation of ALDH4A1 has been linked to metabolic disorders, neurodegenerative conditions and cancer, making it a potential biomarker and therapeutic target.

Performance characteristics: detection range 0.312-20 ng/mL, sensitivity 0.16 ng/mL, intra-assay CV 5.4%, inter-assay CV 8.2%. Linearity in serum (N=5) ranged from 102-112% at 1:2 to 102-113% at 1:16; in EDTA plasma from 103-115% to 97-107%; in heparin plasma from 105-116% to 104-114%. Recovery in serum averaged 104% (range 98-110%) and in plasma averaged 106% (range 100-112%). Synonyms: aldehyde dehydrogenase family 4 member A1, L-glutamate gamma-semialdehyde dehydrogenase, P5C dehydrogenase. Research area: Cell Biology. For research use only.
