Mouse Complement factor D (Cfd) ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of mouse complement factor D in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody sandwich format with a colorimetric readout (TMB substrate, absorbance measured at 450 nm) and is intended for research in immunology, particularly studies of the alternative complement pathway, autoimmune disease and infectious disease.

The kit delivers a sensitivity of 28.125 pg/mL with a working range of 46.875–3000 pg/mL. Reagents are stored at 2–8°C and remain stable for 12 months. Validated linearity across dilutions of 1:2, 1:4 and 1:8 shows serum values of 91–104%, 85–101% and 86–102%; EDTA plasma 86–99%, 82–101% and 84–99%; and heparin plasma 82–95%, 82–95% and 84–95%. Spike-and-recovery performance averages 93% in serum, 100% in EDTA plasma and 93% in heparin plasma.

The workflow involves plate and reagent equilibration, a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-streptavidin binding step, 10–20 minutes of TMB colour development in the dark, and a stop reaction read at 450 nm. Sample preparation guidance covers serum (clot at 1000 × g for 20 minutes), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (2500 rpm for 5 minutes) and cell lysate protocols.
