Description
A sandwich ELISA from Assay Genie designed for the quantitative measurement of Collagen Type X (COL10) in mouse samples. The kit uses a double-antibody sandwich format with colorimetric detection, and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Assay sensitivity is 0.188 ng/mL, with a working range of 0.313–20 ng/mL. Reagents are stored at 2–8°C for 12 months.
Collagen Type X is a key component of the extracellular matrix in bone and cartilage tissues, where it plays a critical role in skeletal development and growth. Abnormal levels of COL10 have been linked to skeletal dysplasias and osteoarthritis, supporting its use as a biomarker for studying these disorders. Synonyms recognised for the target include COL10, COL10A1, Collagen alpha-1(X) chain, and COL10-A1.
Reported linearity for serum (n=5) spans 88–105% at 1:2, 85–103% at 1:4, and 90–102% at 1:8; EDTA plasma (n=5) 82–98%, 83–98%, and 82–99%; and heparin plasma (n=5) 80–95%, 81–100%, and 81–97%. Recovery averages 95% for serum (87–105% range), 94% for EDTA plasma (89–97%), and 96% for heparin plasma (89–103%). The protocol uses a primary incubation at 37°C for 90 minutes, a biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB colour development in the dark for 10–20 minutes, with absorbance read at 450 nm after stop solution is added.
