Description

This colorimetric sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of coagulation factor V in mouse samples. It employs a double-antibody sandwich format using a biotin-labeled detection antibody and HRP-streptavidin conjugate, with signal generation via TMB substrate and absorbance read at 450 nm.

According to the manufacturer, sensitivity is 1.875 ng/mL and the assay covers a range of 3.125–200 ng/mL. The kit is validated for plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with serum and tissue homogenates additionally referenced in product documentation. Storage is at 2–8°C for 12 months. Coagulation factor V is a key component of the blood clotting cascade, facilitating conversion of prothrombin to thrombin, and its dysregulation is implicated in various bleeding disorders, making this kit relevant to hemostasis and coagulation research.

Manufacturer-reported linearity for EDTA plasma (n=5) is 83–97% at 1:4 dilution and 90–100% at 1:8 dilution; heparin plasma (n=5) shows 91–101%, 83–98%, and 87–99% at 1:2, 1:4, and 1:8 dilutions respectively. Recovery is 88–105% (average 98%) in EDTA plasma and 86–102% (average 96%) in heparin plasma. The procedure involves primary incubation at 37°C for 90 minutes, detection antibody binding for 60 minutes, and HRP-streptavidin binding for 30 minutes, followed by 10–20 minutes of substrate development in the dark.
