Mouse CMA1/Chymase ELISA Kit

A colorimetric sandwich ELISA from Assay Genie for the quantitative measurement of mouse chymase (CMA1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Chymase is a mast-cell serine protease involved in tissue remodelling, inflammation and immune regulation, and its dysregulation has been implicated in asthma, fibrosis and cardiovascular disorders, making it a relevant research biomarker. Synonyms include Cma1, Alpha-chymase and Mast cell protease I, and the assay is referenced under UniProt P21844.

The protocol comprises six steps: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding at 37°C for 60 minutes; HRP-streptavidin (SABC) binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and absorbance reading at 450 nm after stop solution. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant and cell lysate. Reported recovery in serum is 89–96% (average 93%), in EDTA plasma 91–102% (average 95%), and in heparin plasma 94–102% (average 98%). Linearity across 1:2, 1:4 and 1:8 dilutions is given for serum (85–98%, 90–104%, 86–105%), EDTA plasma (82–100%, 85–100%, 84–101%) and heparin plasma (84–98%, 82–87%, 82–100%). Components supplied include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-streptavidin conjugate concentrate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers and a technical manual. The kit is stored at 2–8°C for 12 months.
