CKLFSF8, more commonly referenced in the literature as CMTM8 (CKLF-like MARVEL transmembrane domain-containing protein 8), is a transmembrane chemokine-like factor that has been investigated as a putative tumor suppressor in epithelial cancers and as a modulator of lymphocyte and monocyte signaling. The Assay Genie sandwich ELISA pairs a capture antibody pre-immobilized on the microplate with a biotinylated detection antibody and streptavidin-HRP, producing an absorbance signal proportional to CMTM8 concentration that is read at 450 nm, with a reference filter at 570 or 630 nm often used for plate background correction. Reported performance is consistent with a standard curve spanning approximately 0.3 to 20 ng/mL, intra-assay precision below 10% CV and inter-assay precision below 12% CV, supporting quantitative comparisons between treated and untreated mouse cohorts. Serum and plasma samples are typically clarified by centrifugation at 1000 x g for 15 minutes at 2 to 8 degrees C prior to loading, while tissue homogenates and cell lysates require dilution in assay buffer to fall within the dynamic range. Each 96-well plate kit contains pre-coated strip wells, lyophilized recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer concentrate, substrate solution, stop solution and adhesive plate seals sufficient for 96 determinations. Unopened reagents should remain refrigerated at 2 to 8 degrees C and protected from light, while reconstituted standards are best aliquoted and stored frozen at -20 degrees C or below to limit repeated freeze-thaw cycles; reconstituted antibody and streptavidin-HRP working solutions are normally stable only for short term use. The kit is supplied for in vitro research use only and is not intended for diagnostic procedures.
