The assay is validated for linearity across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with values ranging from 80% to 105%. Recovery averages are 95% for serum (range 89-102%), 96% for EDTA plasma (range 87-104%), and 93% for heparin plasma (range 86-101%). The protocol involves a 90-minute primary incubation at 37°C, followed by 60-minute detection antibody binding, 30-minute HRP-streptavidin binding, and TMB color development in the dark for 10-20 minutes, before absorbance is read at 450 nm. Reagents are stored at 2-8°C for 12 months, with standards and plate strips also stable at -20°C for extended storage.
