Description

Designed for the precise quantification of CD137 (also known as TNFRSF9) in mouse samples, this Sandwich ELISA from Assay Genie uses a double-antibody format with a sensitivity of 4.688 pg/mL and a working range of 7.813-500 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. CD137 is a protein involved in immune responses, particularly in regulating T cell activation and proliferation, and its dysregulation has been implicated in various autoimmune diseases and cancer, making it a valuable biomarker for research into these conditions.

The protocol begins with reagent and TMB substrate equilibration to room temperature, followed by loading standards, samples and controls onto the pre-coated plate. A 90-minute primary incubation at 37°C allows antigen binding, after which a biotin-labeled detection antibody is added for 60 minutes at 37°C, then HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added and developed in the dark for 10–20 minutes; the reaction is then stopped and absorbance read at 450 nm. Sample preparation guidance is provided for serum (centrifuge at 1000 × g for 20 minutes), plasma (centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate.

Linearity at 1:2, 1:4 and 1:8 dilutions is 89-103% / 84-99% / 81-93% for serum, 88-103% / 83-100% / 83-99% for EDTA plasma and 87-102% / 86-97% / 85-92% for heparin plasma (n=5 each). Recovery ranges are 88-104% (average 96%) for serum, 89-101% (average 95%) for EDTA plasma and 86-103% (average 95%) for heparin plasma. All components are stored at 2-8°C for 12 months and shipped at +4°C.
