Mouse Cathepsin L ELISA Kit

Assay Genie provides a sandwich ELISA kit designed for the precise measurement of cathepsin L levels in mouse samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Cathepsin L is a key enzyme involved in intracellular protein breakdown, and its dysregulation has been linked to cancer, autoimmune disorders, and infectious diseases, making it a valuable biomarker for investigating these conditions and identifying potential therapeutic targets.

The colorimetric assay has a sensitivity of 18.75 pg/mL and a range of 31.25-2000 pg/mL. Kit components are stored at 2-8°C for 12 months. Linearity across serum, EDTA plasma, and heparin plasma samples (n=5) at 1:2, 1:4, and 1:8 dilutions showed ranges of 88-104%, 85-103%, and 85-104% for serum; 83-101%, 87-95%, and 85-94% for EDTA plasma; and 81-90%, 82-98%, and 81-98% for heparin plasma. Recovery testing showed averages of 98% for serum (range 87-105%), 91% for EDTA plasma (range 87-95%), and 96% for heparin plasma (range 88-101%).

The assay protocol involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution addition.
