CRTAP operates as a key subunit of the collagen prolyl 3-hydroxylation complex, the enzymatic system responsible for 3-hydroxylation of specific proline residues on fibrillar collagens such as types I, II, and V. Mouse Crtap loss-of-function models recapitulate recessive osteogenesis imperfecta type VII, giving investigators a tractable system for studying collagen biosynthesis, skeletal development, craniofacial morphogenesis, and connective tissue disease mechanisms.
