Description

Assay Genie offers a Sandwich ELISA kit for the precise detection of Bone Morphogenetic Protein 1 (BMP1) in mouse samples, using a colorimetric detection method in a double-antibody sandwich format. The kit is intended for research applications in cell biology. BMP1 is involved in biological processes including bone formation, cell differentiation, and tissue regeneration, and its dysregulation has been linked to conditions such as osteogenesis imperfecta and cardiovascular diseases, making it a focus of interest as a potential therapeutic target and diagnostic biomarker.

The assay accepts a broad range of mouse sample types, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported performance characteristics include a sensitivity of 46.875 pg/mL and an assay range of 78.125–5000 pg/mL. Linearity across dilutions of 1:2, 1:4, and 1:8 spans 87–92%, 85–97%, and 85–100% for serum (n = 5); 87–102%, 86–100%, and 86–100% for EDTA plasma (n = 5); and 90–101%, 82–97%, and 87–99% for heparin plasma (n = 5). Spike-and-recovery values span 86–104% across serum, EDTA plasma, and heparin plasma (n = 5), with average recoveries of 92% (serum), 97% (EDTA plasma), and 95% (heparin plasma). Reagents are stored at 2–8°C with a shelf life of 12 months.

The protocol follows a six-step workflow: equilibrate reagents and TMB substrate to room temperature and set up standards, samples, and controls on the pre-coated plate; incubate for 90 minutes at 37°C to allow antigen binding; add biotin-labelled detection antibody and incubate for 60 minutes at 37°C; incubate with HRP-Streptavidin (SABC) for 30 minutes at 37°C; develop with TMB substrate for 10–20 minutes in the dark; and add stop solution before measuring absorbance at 450 nm. Sample preparation guidance includes allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes, and centrifuging anticoagulant-collected plasma at 1000 × g for 15 minutes at 2–8°C.
