Performance data show recovery averages of 94% in serum, 94% in EDTA plasma, and 91% in heparin plasma, with linearity ranging from 82% to 105% across dilution factors of 1:2, 1:4, and 1:8. The assay procedure involves equilibrating reagents and loading the pre-coated plate with standards and samples for 90 minutes at 37°C, followed by biotin-labeled detection antibody (60 minutes at 37°C), HRP-streptavidin binding (30 minutes at 37°C), TMB substrate development in the dark for 10-20 minutes, and absorbance measurement at 450 nm.

Sample preparation includes allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes, collecting plasma from anticoagulant tubes centrifuged at 1000 × g for 15 minutes at 2-8°C, homogenizing tissue in PBS with protease inhibitors, centrifuging cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells with buffer containing protease inhibitors.
