Description

A sandwich ELISA from Assay Genie designed for the detection of mouse BAD (Bcl-2-associated death promoter) in serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The kit uses a double-antibody sandwich format with colorimetric detection, where absorbance is read at 450 nm following TMB substrate development.

BAD is a key protein involved in apoptosis, playing a critical role in regulating cell death, and its dysregulation has been linked to cancer, neurodegenerative disorders, and autoimmune conditions. The assay delivers a sensitivity of 18.75 pg/mL with a measuring range of 31.25–2000 pg/mL. Reported linearity spans 87–104% for serum, 82–100% for EDTA plasma, and 81–99% for heparin plasma across dilution factors. Recovery averages 94% in serum, 96% in EDTA plasma, and 96% in heparin plasma. Reagents should be stored at 2–8°C, with the kit stable for 12 months under these conditions.

The workflow includes equilibrating reagents and loading standards and samples onto the pre-coated plate, primary incubation at 37°C for 90 minutes, addition of biotin-labelled detection antibody with a 60-minute incubation, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm immediately after adding stop solution.
