Apolipoprotein A-II (ApoA-II) is the second most abundant protein component of high-density lipoprotein (HDL) particles and plays a meaningful role in lipid metabolism, HDL remodeling, and reverse cholesterol transport. This sandwich ELISA from Assay Genie uses colorimetric detection and is validated for mouse serum and plasma samples, delivering quantitative measurement of ApoA-II across a broad dynamic range. The 96-well microplate format supports high-throughput studies in cardiovascular, metabolic, and inflammation research, with results typically generated within a few hours. Reagents should be stored refrigerated at 2-8°C, and the kit is intended for research use only.
Each kit contains a pre-coated 96-well microplate, capture antibody, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, diluent, TMB substrate, stop solution, and a recombinant mouse ApoA-II standard calibrated to generate an 8-point standard curve. The assay uses approximately 50 ul of diluted serum or plasma per well, with incubations totalling around 2.5 hours at room temperature, and absorbance is read at 450 nm on a standard plate reader.
Apolipoprotein A-II is synthesized in the liver and circulates as a disulfide-linked homodimer at residue Cys6, predominantly on HDL. The Assay Genie protocol has been validated on C57BL/6, ApoE-/-, and LDLR-/- mouse strains, and is frequently used in studies of PPAR-alpha agonists, LXR ligands, and high-fat diet interventions. Endogenous plasma concentrations in healthy mice typically fall between 30-80 ug/ml, and the working dynamic range is calibrated to cover both physiological and elevated pathological levels.
No significant cross-reactivity has been observed with ApoA-I, ApoC-III, or ApoE. All components should be stored at 2-8°C, with the standard aliquoted and frozen at -20°C if the kit will not be used within 2 weeks. Each pack is for research use only and is not intended for diagnostic procedures.
