Mouse Anti-Thyroid-Peroxidase Antibody/TPO-Ab ELISA Kit

Designed for the quantitative detection of thyroid peroxidase (TPO) antibodies in mouse samples, this Sandwich ELISA kit from Assay Genie uses a double-antigen format with colorimetric readout. TPO is an enzyme essential for thyroid hormone synthesis, and TPO antibodies are often elevated in autoimmune thyroid diseases such as Hashimoto's thyroiditis and Graves' disease, making the kit a useful tool for research into these disorders.

The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a reported sensitivity of 0.938 IU/mL and an operating range of 1.563–100 IU/mL. Linearity is reported for serum (1:2 at 85–101%, 1:4 at 81–101%, 1:8 at 83–100%), EDTA plasma (1:2 at 83–94%, 1:4 at 81–101%, 1:8 at 80–96%) and heparin plasma (1:2 at 89–99%, 1:4 at 84–97%, 1:8 at 82–96%). Recovery is reported as 85–96% (average 91%) in serum, 85–94% (average 91%) in EDTA plasma, and 86–99% (average 95%) in heparin plasma. Reagents should be stored sealed at 2–8°C and not cryopreserved.

Following reagent equilibration to room temperature, standards, samples, and controls are loaded onto the pre-coated plate and incubated at 37°C for 90 minutes. A biotin-labeled antigen is then added for a 60-minute incubation, followed by HRP-streptavidin for 30 minutes. TMB substrate is developed in the dark for 10–20 minutes, the reaction is stopped, and absorbance is read at 450 nm.
