Alpha1-microglobulin (A1M, also referred to as AMBP or protein HC) is encoded by the AMBP gene and translated as a precursor that is later cleaved into the circulating ~26 kDa lipocalin subunit and the Kunitz-type protease inhibitor bikunin. In circulation the protein is found mostly as a monomer and as a high-molecular-weight complex with IgA, and it exerts broad radical-trapping and heme-binding antioxidant activity in addition to its role as a tubular reabsorption marker. The Assay Genie chemiluminescent immunoassay therefore complements classical urinary injury readouts such as NGAL, KIM-1, and beta-NAG, allowing researchers to stratify glomerular filtration versus proximal tubular reabsorption in the same animal. The 96-well microplate is pre-blocked and coated with a capture antibody specific for mouse alpha1-MG, and the supplied lyophilized standards are reconstituted in assay buffer to generate a seven-point calibration curve (typically spanning ~0.5 to 100 ng/mL). Serum and plasma samples are generally diluted 1:50 to 1:200 in sample diluent, while urine is usually assayed neat or at a 1:5 dilution depending on diuresis; all samples should be clarified by centrifugation at 1,000 x g for 10 minutes at 4 °C and stored aliquoted at -80 °C to limit freeze-thaw cycles. The detection reagent is an acridinium-ester-labelled antibody that emits light upon trigger addition, read on a luminometer with a broad dynamic range and an analytical sensitivity below 1 ng/mL. Each kit ships with a sealed foil pouch of pre-coated wells, lyophilized calibrators, conjugated detection antibody, concentrated wash buffer, substrate trigger solution, and a manual with recommended incubation times of 60 minutes at room temperature for both the primary capture and detection steps. Researchers commonly deploy this panel in cisplatin- and aristolochic-acid-induced nephrotoxicity models, unilateral ureteral obstruction, polycystic kidney disease cohorts, and sepsis challenge studies, and have used it to monitor tubular recovery after ischemic reperfusion injury. Reagents remain stable for 12 months from manufacture when refrigerated at 2-8 °C and protected from light; reconstituted standards should be used within 14 days if kept at 2-8 °C or frozen in single-use aliquots for longer term storage. The assay is intended for in vitro research use only and is not approved for diagnostic or veterinary use.
