The soluble adenylate cyclase ADCY10 is a bicarbonate- and calcium-sensitive enzyme that produces cAMP in compartments distinct from those served by transmembrane adenylyl cyclases, making it a distinct target for cAMP research. Researchers studying male germ cell signaling, airway ciliary beat regulation, or metabolic cAMP microdomains use mouse ADCY10 ELISA kits to quantify protein levels in tissue homogenates and cell lysates. This Assay Genie kit targets the soluble isoform specifically, allowing it to be measured independently of the G protein-coupled transmembrane adenylate cyclases (ADCY1-9) that dominate most cAMP studies. The kit provides reagents for 96 assays and is intended for research use only in mouse-derived samples, supporting projects in reproductive biology, renal physiology, and intracellular pH or bicarbonate signaling. Each kit is supplied with pre-coated plates, detection antibody, standards, and the buffers needed to generate a standard curve, following a protocol suitable for end-point measurement on a standard microplate reader. The assay performs well on freshly prepared homogenates and on frozen aliquots stored at -80 °C; repeated freeze-thaw cycles should be avoided to preserve epitope integrity. For best results, tissues should be homogenized in cold lysis buffer supplemented with protease inhibitors, clarified by centrifugation, and either assayed the same working day or frozen immediately in single-use aliquots for later analysis. Researchers working on soluble adenylyl cyclase knockouts, bicarbonate-sensitive cAMP pools, or sperm capacitation models will find this reagent useful for tracking ADCY10 protein abundance alongside functional cAMP readouts.
