Acetyl-CoA carboxylase alpha (ACACa), also called ACC1, sits at the entry point of de novo fatty acid synthesis by converting acetyl-CoA to malonyl-CoA. Research on hepatic and adipose lipid handling, insulin signaling cascades, AMPK-mediated phosphorylation at Ser79, and tumor lipid metabolism frequently relies on quantifying ACACa protein levels in mouse tissues and cell-derived samples. Assay Genie supplies this quantitative immunoassay configured as a complete kit containing reagents for 96 individual determinations. Researchers apply tissue homogenates and cell lysates to wells pre-coated with capture antibody specific for mouse ACACa. After incubation and washing, a detection antibody conjugated to reporter enzyme binds the captured target, and substrate conversion yields a measurable color change proportional to ACACa concentration in each sample. Each kit ships with a strip plate, lyophilized recombinant standards, diluted detection antibody, wash buffer concentrate, sample diluent, substrate solution, and stop reagent. Standard curve generation using the supplied calibrators allows interpolation of unknown sample concentrations across the working range. Storage at 4 °C preserves reagent activity through the stated shelf life; reconstituted standards should be aliquoted and stored frozen to limit repeated freeze-thaw cycles that compromise protein integrity. The assay is configured for research use only and is not intended for diagnostic procedures. Compatible downstream confirmatory methods commonly include western blot analysis of phosphorylated ACC at Ser79 to corroborate enzyme activity state, and quantitative PCR measurement of ACACA transcript in matching tissue samples.
