Mouse 12-LO (Arachidonate 12-Lipoxygenase) ELISA Kit (MOES01636)

Arachidonate 12-lipoxygenase (ALOX12) is a non-heme iron-containing enzyme that catalyzes the oxygenation of arachidonic acid to 12-hydroperoxyeicosatetraenoic acid (12-HPETE), an upstream intermediate in the biosynthesis of pro-inflammatory lipid mediators. The Mouse 12-LO ELISA Kit from Assay Genie provides a quantitative immunoassay for measuring endogenous 12-LO levels in murine experimental samples and is widely used in cardiovascular, dermatological, and inflammation research.

Reagents are supplied for 96 individual determinations, and the assay is configured for research use only. Investigators apply this kit to monitor lipoxygenase pathway activity in mouse models of atherosclerosis, hepatic steatosis, and chronic inflammatory disease; to evaluate pharmacological inhibitors targeting the 12-LO enzyme; and to characterize transgenic or knockout mouse lines with genetically modified 12-LO expression. Compatible sample matrices include tissue homogenates, cell lysates, and conditioned media from murine cell culture systems.

Capture antibodies specific for mouse 12-LO are pre-coated on the plate. Sequential incubation with standards and experimental samples is followed by addition of a biotinylated detection antibody, streptavidin-HRP conjugate, and a chromogenic substrate. The intensity of the resulting color is proportional to 12-LO concentration, and absolute quantification is achieved by interpolation against a standard curve generated with recombinant mouse 12-LO protein.
