Description

A colorimetric Sandwich ELISA from Assay Genie designed for the quantitative measurement of TARC (Thymus Activation Regulated Chemokine) in monkey serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The double-antibody format reports a sensitivity of 18.75 pg/mL with a working range of 31.25-2000 pg/mL, and unopened kits are stored at 2-8°C for 12 months.

Linearity assessed on spiked serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions returned 87-101%, 82-100% and 85-93% for serum; 85-105%, 82-93% and 83-100% for EDTA plasma; and 85-101%, 84-98% and 84-98% for heparin plasma. Spike-and-recovery experiments gave ranges of 92-104% (average 97%) for serum, 87-105% (average 97%) for EDTA plasma and 85-98% (average 91%) for heparin plasma.

The protocol proceeds in six stages: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and stop reaction with absorbance read at 450 nm. Sample preparation notes include clotting and centrifugation at 1000 x g for 20 minutes for serum, anticoagulant collection followed by centrifugation at 1000 x g for 15 minutes at 2-8°C for plasma, homogenisation in PBS with protease inhibitors for tissue, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and lysis with protease inhibitor buffer for cell lysates.
