Description

Assay Genie supplies a Sandwich ELISA kit employing a double-antibody format for the quantitative measurement of AMBP (Alpha-1-Microglobulin/Bikunin Precursor) in monkey samples. The assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and the kit is intended to be stored at 2–8°C for 12 months.

Performance characteristics reported by the manufacturer include a sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Linearity across dilution has been demonstrated in serum (n = 5) at 1:2 = 95–101%, 1:4 = 87–102% and 1:8 = 99–104%; in EDTA plasma (n = 5) at 1:2 = 82–98%, 1:4 = 84–100% and 1:8 = 82–100%; and in heparin plasma (n = 5) at 1:2 = 84–98%, 1:4 = 91–94% and 1:8 = 81–99%. Spike-and-recovery values are reported as serum (n = 5) range 86–102% with an average of 96%, EDTA plasma (n = 5) range 95–106% with an average of 93%, and heparin plasma (n = 5) range 85–102% with an average of 96%.

The workflow follows a sandwich ELISA procedure: equilibrate reagents and TMB substrate, load standards, samples and a zero control on the pre-coated plate and incubate at 37°C for 90 minutes; incubate with a biotin-labelled detection antibody at 37°C for 60 minutes; incubate with HRP-Streptavidin (SABC) at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; and stop the reaction before reading absorbance at 450 nm. Serum is prepared by clotting and centrifuging at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C. Sample handling guidance for tissue homogenate, cell culture supernatant and cell lysate is also provided.
