Monkey MIP-3a/CCL20 ELISA Kit

The assay uses a colorimetric, double-antibody sandwich format and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 18.75 pg/mL and the assay range is 31.25-2000 pg/mL. Kits should be stored at 2-8°C for 12 months.

Linearity in serum (n=5) was 82-99% at 1:2, 80-93% at 1:4 and 88-100% at 1:8; EDTA plasma (n=5) gave 81-92%, 84-99% and 86-103%; heparin plasma (n=5) gave 86-103%, 94-99% and 88-99%. Recovery ranged from 85-99% (average 94%) in serum, 89-99% (average 95%) in EDTA plasma and 89-103% (average 98%) in heparin plasma. The protocol involves equilibrating reagents and TMB substrate, loading standards, samples and blanks and incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody and incubating at 37°C for 60 minutes, adding HRP-Streptavidin (SABC) and incubating at 37°C for 30 minutes, developing colour with TMB substrate in the dark for 10-20 minutes, then adding stop solution and reading absorbance at 450 nm.
