Monkey IL-12 ELISA Kit

From Assay Genie, this colorimetric Sandwich ELISA, Double Antibody kit enables quantitative measurement of the cytokine IL-12 in monkey samples. The analyte, also known by the synonyms NKSF, CLMF, Interleukin 12, Natural Killer Cell Stimulatory Factor, and Cytotoxic Lymphocyte MatuRation Factor, is detected with a sensitivity of 9.375 pg/mL across an assay range of 15.625-1000 pg/mL. Accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2-8°C for 12 months.

The assay workflow proceeds as follows: equilibrate reagents and TMB substrate to room temperature and assign standard, test sample and control (zero) wells on the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; add TMB substrate and incubate in the dark for 10–20 minutes; add stop solution and read absorbance at 450 nm immediately. Sample preparation guidance from Assay Genie: for serum, allow blood to clot, centrifuge at 1000 × g for 20 minutes and collect supernatant; for plasma, collect using anticoagulant tubes and centrifuge at 1000 × g for 15 minutes at 2–8°C; for tissue homogenate, homogenize in PBS with protease inhibitors, then centrifuge and collect supernatant; for cell culture supernatant, centrifuge at 2500 rpm for 5 minutes and collect clarified supernatant; for cell lysates, lyse cells using lysis buffer with protease inhibitors, centrifuge and collect the protein supernatant.

Reported linearity at 1:2, 1:4 and 1:8 dilutions is: Serum (n = 5) 86-102%, 84-98%, 80-94%; EDTA Plasma (n = 5) 89-105%, 82-101%, 80-98%; Heparin Plasma (n = 5) 93-104%, 83-96%, 80-93%. Reported recovery is: Serum (n = 5) 85-101% range with 94% average; EDTA Plasma (n = 5) 86-105% range with 98% average; Heparin Plasma (n = 5) 88-105% range with 95% average.
