Monkey cTn-I/TNNI3/cardiac Troponin I ELISA Kit

A sandwich double-antibody ELISA from Assay Genie enables quantitative measurement of Monkey cTn-I/TNNI3 (cardiac Troponin I) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The microplate is pre-coated with a capture antibody specific to the target protein; standards and diluted samples bind to the immobilized antibody, after which a biotinylated detection antibody directed against a different epitope forms a sandwich complex. HRP-conjugated Streptavidin and TMB substrate generate a colorimetric reaction that is stopped with an acidic solution and read at 450 nm, with signal intensity directly proportional to analyte concentration as determined from a standard curve.

The assay has a sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL. Kits are stored at 2-8°C for 12 months. Linearity across dilution has been demonstrated in Serum (n=5) at 85-103% (1:2), 91-105% (1:4) and 89-104% (1:8); in EDTA Plasma (n=5) at 88-98% (1:2), 87-101% (1:4) and 89-101% (1:8); and in Heparin Plasma (n=5) at 81-99% (1:2), 82-98% (1:4) and 80-98% (1:8). Recovery values are 86-104% with an average of 99% in Serum (n=5), 88-105% averaging 97% in EDTA Plasma (n=5), and 86-102% averaging 93% in Heparin Plasma (n=5).

The procedure covers reagent equilibration and plate setup, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB color development in the dark for 10-20 minutes, and stop solution addition followed by immediate absorbance measurement at 450 nm.
