Moesin/Ezrin/Radixin Colorimetric Cell-Based ELISA Kit

A Cell-Based ELISA Kit from Assay Genie designed to detect Moesin, Ezrin and Radixin protein expression in cultured adherent cells. These three proteins belong to the ERM family, which links the plasma membrane to the actin cytoskeleton and plays roles in cell structure, membrane dynamics and cell signalling. Dysregulation of ERM proteins has been implicated in cancer, inflammatory disorders and neurological conditions, making them valuable targets for studies of cell biology, disease mechanisms and potential therapeutics.

The kit uses an indirect ELISA format: target proteins are captured by specific primary antibodies, and HRP-conjugated secondary antibodies that bind the Fc region of the primary antibody catalyse a colorimetric reaction upon substrate addition. Detection is read at 450 nm, and the assay is reactive with human, mouse and rat samples. It is lysate-free, allowing relative quantitation of Moesin/Ezrin/Radixin in intact cells as well as screening for the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on target expression. The dynamic range is greater than 5,000 cells.

Two normalisation methods are provided: a monoclonal anti-GAPDH antibody as an internal positive control, and Crystal Violet whole-cell staining for cell density. The kit is supplied with a 96-well cell culture clear-bottom microplate, 10X TBS, quenching buffer, blocking buffer, 15X wash buffer, primary antibody diluent, 100x anti-phospho target antibody, 100x anti-target antibody, anti-GAPDH antibody, HRP-conjugated anti-rabbit IgG antibody, HRP-conjugated anti-mouse IgG antibody, SDS solution, stop solution, ready-to-use substrate, crystal violet solution and adhesive plate seals. Storage is at 4 °C for 6 months.
