Description

From Assay Genie, this cell-based ELISA kit is designed for the measurement of MNK1 phosphorylation at threonine 385 in cell lysates. MNK1 is a key player in the MAPK signaling pathway, regulating cellular processes such as gene expression, cell growth, and survival, and phosphorylation at Thr385 is critical for its activation and function.

The kit uses a colorimetric detection method read at 450 nm and is reactive with human, mouse, and rat samples. It is a lysate-free, high-throughput assay in an indirect ELISA format, where MNK1 (Phospho-Thr385) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

Developed for signal transduction research, the kit supports measurement of relative phosphorylated MNK1 levels in cultured cells as well as screening of treatments, inhibitors (such as siRNA or chemicals), or activators on MNK1 phosphorylation. The target gene is MKNK1 (Gene ID: 8569; UniProt: Q9BUB5; OMIM: 606724), encoding the MAP kinase-interacting serine/threonine-protein kinase 1 with a reported molecular weight of 39,104 Da. The stated dynamic range is > 5000 Cells.
