Description

Assay Genie supplies a cell-based ELISA designed for the qualitative measurement of MMP-16 protein expression in cultured cells. The assay uses an indirect ELISA format, in which MMP-16-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. Because the readout is qualitative, two normalization strategies are included: a monoclonal anti-human GAPDH antibody for internal positive control, and a Crystal Violet whole-cell staining step to determine cell density so absorbance values can be adjusted for plating differences.

The kit is configured for use with adherent cells and is intended for measuring relative MMP-16 levels as well as screening the effects of treatments, inhibitors (including siRNA or chemicals), or activators. It is reactive with human, mouse and rat samples, employs a colorimetric detection method read at 450 nm, and has a stated dynamic range of > 5000 Cells. Storage condition is 4°C with a stability of 6 Months. MMP-16 (matrix metalloproteinase-16) is a zinc-dependent endopeptidase of the M10A family that degrades extracellular matrix components such as collagen type III and fibronectin and activates progelatinase A, making it a relevant marker in studies of matrix remodeling, cell migration, cancer, inflammation and tissue fibrosis.
