Description

From Assay Genie, this Cell-Based ELISA Kit provides a sensitive, lysate-free, high-throughput colorimetric assay for measuring MMP-13 protein expression directly in cultured cells. Designed for use with adherent cell samples, the kit is supplied in a 96-Well Microplate format, uses a Colorimetric detection method read at 450 nm, and has a stated dynamic range of greater than 5000 cells. The product is intended for research in cell biology with a reported reactivity against human samples and is stored at 4°C with a stated stability of 6 months.

Detection follows an indirect ELISA workflow: target protein is captured by MMP-13-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of those primaries to catalyse a colorimetric reaction on substrate addition. The kit includes a monoclonal anti-human GAPDH antibody as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step for determining cell density so that absorbance readings can be adjusted to cell number. This allows measurement of the relative amounts of MMP-13 in cultured cells and supports screening of treatments, inhibitors such as siRNA or chemicals, or activators on MMP-13 expression.

MMP-13, also known as collagenase-3, is a matrix metalloproteinase that degrades extracellular matrix proteins and plays a critical role in tissue remodelling and wound healing. Dysregulation of MMP-13 has been implicated in pathological conditions including arthritis, cancer and cardiovascular disease, making the target of interest for studies of disease mechanisms and potential interventions. Researchers can use the kit to explore the role of MMP-13 in both physiological and pathological processes at the cellular level.
